Thymosin Alpha-1 (Tα1) is a 28-amino-acid, N-terminally acetylated peptide originally isolated from thymosin fraction 5, a calf thymus extract. Research overview
Tα1 was isolated and sequenced by Allan Goldstein’s group in the 1970s. It comes from the N-terminus of prothymosin α. Mechanistic studies point to the innate immune system: a 2004 study in Blood showed that Tα1 activates dendritic cells through Toll-like receptor signalling (including the MyD88-dependent pathway), with downstream effects on Th1 responses in murine models [1].
In-vitro work with human cells has refined that picture. A 2015 study examined how Tα1 affects human monocyte-derived dendritic cells stimulated with viral and bacterial TLR agonists, and described context-dependent effects on cytokine production [2].
A 2016 review summarises Tα1’s immunomodulatory mechanisms, including its effects on T-cell maturation, dendritic-cell function and cytokine networks [3]. In the laboratory, Tα1 is used in dendritic-cell and T-cell culture assays, TLR signalling studies and as a reference thymic peptide.

Storage and handling
Thymosin Alpha-1 ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of Thymosin Alpha-1 at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
- Handle with gloves and eye protection, following your laboratory’s standard chemical-hygiene practice.
