Research overview
Oxyntomodulin is a natural proglucagon-derived peptide that weakly activates both GLP-1R and GCGR. Mazdutide was designed as a long-acting oxyntomodulin-like analogue. Its sequence is modified to balance activity at the two receptors, and a fatty-acid side chain binds albumin to extend its half-life. Both receptors are class B GPCRs that signal mainly through Gαs and cAMP, but they differ in tissue distribution and downstream biology.
A 2025 review in Drugs summarises Mazdutide’s structure, receptor pharmacology and development history [1]. In preclinical research, a 2025 study in EBioMedicine combined mouse models with multi-omics analysis to examine GLP-1R/GCGR dual agonism in the brain [2].
In vitro, Mazdutide is profiled in GLP-1R- and GCGR-expressing cell lines to compare cAMP potency and signalling bias at each receptor. It is also used as a reference standard for LC-MS identification and peptide-stability methods.
