Research overview
In 2001, Kotani and colleagues showed that peptides encoded by the metastasis-suppressor gene KiSS-1, which they named kisspeptins, are the natural ligands of the orphan G protein-coupled receptor GPR54. Kisspeptin-10 activated the receptor with potency similar to longer forms [1]. KISS1R couples mainly through Gαq/11, leading to phospholipase C activation and intracellular calcium mobilisation.
Genetic studies later placed kisspeptin–GPR54 signalling at the top of the hypothalamic-pituitary-gonadal axis, and it became one of the most active areas of reproductive neuroendocrinology. Seminara reviews the discovery of this pathway [2].
Kisspeptin-10 is also studied outside the reproductive axis. A 2018 study reported that it stimulated osteoblast differentiation in vitro through GPR54-mediated regulation of BMP2 [3]. Laboratory uses include KISS1R binding and calcium assays, GnRH-neuron and cell-line studies and structure–activity work on kisspeptin analogues.

Storage and handling
Kisspeptin-10 ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of Kisspeptin-10 at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
