Research overview
Tri-agonist peptides combine sequence elements of glucagon, GLP-1 and GIP in one molecule so that a single ligand activates all three receptors. Finan and colleagues described the rational design of a monomeric peptide triagonist in 2015, including how they tuned the relative potency at each receptor through iterative sequence changes [1].
Medicinal chemists have refined the concept since. A 2022 Molecular Metabolism study described next-generation GLP-1/GIP/glucagon triple agonists and characterised their receptor potency in cell-based assays alongside studies in mice [2]. The same year, Coskun and colleagues reported the discovery and pharmacology of a fatty-acid-acylated tri-agonist, including in-vitro cAMP potency at each human receptor and receptor-selectivity profiling [3].
In the laboratory, tri-agonist peptides are profiled in GIPR-, GLP-1R- and GCGR-expressing cell lines using cAMP accumulation, β-arrestin recruitment and receptor-internalisation assays. They also serve as reference standards for LC-MS and peptide-stability method development.

Storage and handling
GLP-3 (RET) ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of GLP-3 (RET) at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
- Handle with gloves and eye protection, following your laboratory’s standard chemical-hygiene practice.
