GHRP-6 (Growth Hormone Releasing Hexapeptide) is a synthetic six-residue peptide, His-D-Trp-Ala-Trp-D-Phe-Lys-NH2, and one of the first synthetic agonists of the receptor later identified as the ghrelin receptor (GHS-R1a). Research overview
GHRP-6 was described in 1984 by Bowers and colleagues, who showed that the hexapeptide acts directly on the pituitary to release growth hormone in both in-vitro and in-vivo models [1]. Its activity at an unknown receptor led eventually to the cloning of the growth hormone secretagogue receptor and the discovery of ghrelin as its natural ligand.
Later in-vitro studies compared GHRP-6 with GHRP-2 in rat primary pituitary cells [2] and measured intracellular cAMP alongside GH secretion in ovine and rat pituitary cells. These experiments helped show that GHRPs signal differently from GHRH [3].
Today GHRP-6 is used as a reference GHS-R1a agonist in binding and calcium-mobilisation assays, as a comparator for newer secretagogues and as an analytical standard.

Storage and handling
GHRP-6 ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of GHRP-6 at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
- Handle with gloves and eye protection, following your laboratory’s standard chemical-hygiene practice.
