Research overview
NAD+ and its reduced form NADH carry electrons in glycolysis, the TCA cycle and oxidative phosphorylation. NAD+ is also consumed as a substrate by signalling enzymes: sirtuin deacylases, poly(ADP-ribose) polymerases (PARPs) and the NADase CD38. Cells maintain their NAD+ pool through de novo synthesis from tryptophan and through salvage pathways using nicotinamide, nicotinamide riboside and nicotinamide mononucleotide. Covarrubias and colleagues review these pathways and how NAD+ levels change in cellular and animal models [1].
A widely cited 2015 review in Science summarises how NAD+-consuming enzymes link cellular energy status to transcription, DNA repair and mitochondrial function in experimental systems [2].
In the laboratory, NAD+ is used as a cofactor in dehydrogenase and sirtuin activity assays, as a substrate for PARP and CD38 enzymology, as a calibration standard for NAD+/NADH quantification by enzymatic cycling or LC-MS, and as a supplement in cell-culture metabolism experiments.

Storage and handling
NAD+ ships as a lyophilized (freeze-dried) solid. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of NAD+ at −20 °C, dry and protected from light. This material is hygroscopic, so keep the vial tightly closed and let it reach room temperature before opening to prevent moisture uptake.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
- Handle with gloves and eye protection, following your laboratory’s standard chemical-hygiene practice.
