Research overview

NNMT transfers a methyl group from S-adenosylmethionine to nicotinamide, producing 1-methylnicotinamide. It therefore sits at the junction of the methionine cycle and the NAD+ salvage pathway. In 2017, Neelakantan and colleagues published a structure–activity study of small-molecule NNMT inhibitors and identified quinolinium scaffolds, including 5-amino-1-methylquinolinium, as potent and selective inhibitors [1].

Follow-up work showed that these compounds cross cell membranes, inhibit NNMT inside cells, raise intracellular NAD+ and S-adenosylmethionine, and alter lipogenic gene expression in cultured adipocytes, with supporting studies in diet-induced mouse models [2]. A 2019 study looked at how NNMT inhibition affects the activation of senescent muscle stem cells in aged mice [3].

In the laboratory, 5-Amino-1MQ is used as a reference NNMT inhibitor in enzyme assays, for studying methylation and NAD+ flux in cell lines and as a comparator in medicinal-chemistry programmes.

Storage and handling

5-Amino-1MQ ships as a lyophilized (freeze-dried) solid. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.

• Store unopened vials of 5-Amino-1MQ at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
• Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
• For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
