Research overview
GIP and GLP-1 are the two main incretin hormones. Each signals through its own class B G protein-coupled receptor, GIPR and GLP-1R, and both couple mainly to Gαs and cAMP. Unimolecular dual agonists combine residues from both hormones in a single peptide chain so that one molecule engages both receptors. Finan and colleagues described the design principles of such dual incretin peptides in 2013 [1].
A 2023 review in Cell Metabolism covers the chemistry, receptor pharmacology and physiology of GIPR/GLP-1R dual agonists. It discusses balanced versus imbalanced receptor potency, biased signalling at GLP-1R (reduced β-arrestin recruitment and internalisation) and how fatty-acid acylation extends half-life through albumin binding [2].
For background on the GLP-1R side of the pharmacology, Drucker’s 2018 review is a standard reference [3]. In vitro, dual agonists are profiled in GIPR- and GLP-1R-expressing cell lines with cAMP, β-arrestin and internalisation assays, and are used as reference standards in LC-MS method development.

Storage and handling
GLP-2 (TIR) ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of GLP-2 (TIR) at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
- Handle with gloves and eye protection, following your laboratory’s standard chemical-hygiene practice.
