Research overview
Native GHRH(1–44) is cleaved quickly between residues 2 and 3 by DPP-4, which inactivates it. Tesamorelin, originally coded TH9507, keeps the full native sequence and adds a trans-3-hexenoic acid to the N-terminus. This protects the peptide from cleavage while preserving its affinity for the GHRH receptor, a Gαs-coupled class B GPCR on pituitary somatotrophs.
The preclinical pharmacology and safety programme for TH9507 was published in 2007. It described receptor activity, stimulation of growth-hormone release in animal models and toxicology findings [1]. Another study in dogs examined pulmonary delivery of the peptide as part of formulation research [2]. A 2006 drug-evaluation review summarises its chemistry and pharmacological profile [3].
In the laboratory, Tesamorelin is used as a stabilised reference GHRH receptor agonist in pituitary-cell cAMP and secretion assays, in peptide-stability studies comparing modified and native GHRH, and as an analytical standard for LC-MS detection of GHRH analogues.

Storage and handling
Tesamorelin ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
- Store unopened vials of Tesamorelin at −20 °C, dry and protected from light. Let the vial reach room temperature before opening to limit condensation.
- Prepare stock solutions in a solvent suited to your assay, label them with concentration and date, and keep them at 2–8 °C for short-term use.
- For longer storage of solutions, divide them into single-use aliquots and freeze them; avoid repeated freeze–thaw cycles.
