Using a reconstitution solution
Lyophilized (freeze-dried) peptides are stored dry because most peptides degrade much faster in solution, through hydrolysis, oxidation, deamidation and aggregation. Reconstitution is the step where a researcher adds a measured volume of solvent to the dry cake to make a stock solution of known concentration. Solvent choice affects how stable the solution is. Pure water suits many short peptides. Some laboratories prefer a preserved aqueous diluent when they plan to draw from a stock vial more than once. Hydrophobic sequences may need a small amount of organic co-solvent or a pH adjustment, as the peptide’s data sheet specifies. Good practice is to let both the peptide vial and the solvent reach room temperature, add the solvent slowly down the vial wall, swirl gently rather than shake, label the vial with its concentration and date, and store the stock as your protocol directs, usually at 2–8 °C.
