Research overview
Amylin is a 37-residue peptide co-secreted with insulin. It signals through amylin receptors (AMY1–3), which form when the calcitonin receptor pairs with one of the receptor activity-modifying proteins RAMP1, RAMP2 or RAMP3. Hay and colleagues’ review in Pharmacological Reviews is the standard reference on amylin receptor composition, ligand selectivity and signalling [1].
Native amylin aggregates and fibrillates readily, which makes it hard to work with in solution. The 2021 Journal of Medicinal Chemistry paper describing cagrilintide’s development explains how researchers addressed this. They substituted residues to reduce fibrillation, added an N-terminal C20 diacid lipid side chain to bind albumin, and characterised the analogue’s potency at amylin and calcitonin receptors in cell-based assays [2].
Cagrilintide is used in vitro as a reference agonist for AMY receptor subtypes and the calcitonin receptor, in cAMP signalling assays and in peptide-stability or LC-MS method development work.

Cagrilintide ships as a lyophilized (freeze-dried) powder. Dry material is far more stable than material in solution, so keep it in its dry, sealed state until you need it.
